About THERAHEAL PN

THERAHEAL PN consists of thermally stabilized natural High and Low molecular weight Hyaluronic acid (H/L-HA) hybrid complex and Polynucleotide (PN).
H/L-HA combines H-HA and L-HA to obtain injectable concentrations of H/L hybrid complex HA. Hyaluronic acids are distributed widely throughout connective, epithelial, and neural tissues. As one of the chief components of the extracellular matrix, it contributes significantly to cell proliferation and migration.THERAHEAL PN not only boosts and hydrates the skin, but also remodels aging and sagging tissue.
PN is a biological composite material that has the biological functions of deoxyribonucleic acid (DNA) and ribonucleic acid (RNA). PDRN(Polydeoxyribonucleotide) is a polymerized molecule of deoxyribonucleotides derived from PN. PN is higher molecule than PDRN. It is a colorless and transparent liquid form in the form of a gel that maintains a constant viscosity, and when injected into the skin, it gradually breaks down while maintaining a constant form in the body. It contributes to tissue recovery, wound healing, and anti-aging effects.

Effect of THERAHEAL PN

By injecting the components that create cells directly into the skin, it promotes the cell's own regenerative ability.
By improving the dermis layer that has become thinner due to aging and damage, it increases the density and thickness of the epidermis and dermis, and pores are also improved, helping to improve skin elasticity and skin texture.
It helps restore normal skin damaged by UV rays and skin irritation.
As the product is injected into the skin, the skin's own epidermis or dermis fills up, resulting in the improvement of fine wrinkles.

Recommended Area of THERAHEAL PN Treatment

It is effective for wrinkles caused by decreased collagen in the dermal layer of the skin, especially around the eyes, where wrinkles are more likely to form due to thin skin among facial wrinkles. By improving the dermis layer that has become thinner due to aging and damage, the density of the dermis and pores are also improved, which helps improve skin texture if treated consistently. Additionally, this can be used not only on the face, but also on the body, such as improving wrinkles on the neck, arms, knees, and abdomen.

Treatment Technique

1) Face

THERAHEAL PN only needs 5 points per cheek for treatment of the whole face unlike other skin boosters. In total, there are 10 injection points for the entire face.
Each treatment requires 2~2.5ml of THERAHEAL PN allowing for maximal diffusion of The hybrid HA complex and PN across the entire face.
10 Aesthetic Points for face (F10-AP) are specially selected areas of the face that have no large blood vessels or nerve branches, thereby minimizing the risk of bruising and improving safety.
With only 5 injection points per cheek, discomfort and downtime is minimal to none.

2) Neck

THERAHEAL PN for neck rejuvenation is excellent for targeting wrinkled or sagging neck skin.
points along the neck (10 Aesthetic Points for Neck: N10-AP) are specially chosen to avoid vital structures such as blood vessels.
Neck anti-aging benefits are stimulated from deep within the skin.
The treatment experience is similar to THERAHEAL PN for the face.

Number of Uses and Capacity

Depending on individual skin needs, we generally recommend treatments every 3 weeks for a total of 3-4 sessions. The results typically last around 6 to 8 months.
For targeted areas like the cheeks, a suggested dosage is 2ml, whereas for a full-face treatment, 4ml is recommended. However, dosages may vary based on facial surface area, so please take this as a reference.

Precautions After the Procedure

Since the THERAHEAL PN components are injected into dermal skin by injection, there are bruises and swelling like normal injection procedures, but there are no side effects such as necrosis, turn-off, or skin rejection.
You should avoid irritating places such as saunas for a week. It may partially bruise or swell, but it disappears quickly. And avoid drinking alcohol, quitting smoking and exercising excessively for a week after the procedure.

Non-recommended Person

1) Those who want immediate effects like Botox, fillers, or lifting procedures :
This treatment improves the overall skin condition by injecting the regenerative ingredients of THERAHEAL PN, so if you are expecting the same effects as Botox or fillers, it may not be for you.
2) Those who want immediate results in shrinking pores or improving pigmentation :
THERAHEAL PN may be possible as an adjuvant treatment, but it is difficult to see results with THERAHEAL PN alone.

PN (Polynucleotide) &
PDRN (Polydeoxyribonucleotide)

Anti-Aging & Repair

PDRN(Polydeoxyribonucleotide) is a polymerized molecule of deoxyribonucleotides derived from a controlled purification process of Salmon Trout or Chum Salmon sperm DNA which makes sure its safety and stability. PDRN activates the adenosine A2A receptors and generates nucleotides that can contribute to DNA formation, thus having a tissue repairing, wound healing and anti-aging effect. Therefore, it is widely used in mesotherapy and dermal fillers. PN (Polynucleotide) is also obtained by extracting sperm DNA from Salmon Trout or Chum Salmon, and higher molecule than PDRN (Polydeoxyribonucleotide). It stimulates collagen production, enhancing skin elasticity and reducing wrinkles.

Mechanism of Action
※ The mechanism of action of PN is almost the same as PDRN.

1) Activation of Adenosine A Receptor

Adenosine activates four distinct adenosine receptors indicated as A1, A2A, A2B, and A3, the A2A receptor plays a central role in modulating inflammation, oxygen consumption, ischemia, cell growth, and angiogenesis. PDRN is able to generate active deoxyribonucleotides, nucleosides, and bases exerting their pharmacological effects and activating the A2A receptor, PDRN interacting with the A2A receptor provides a more complete tissue regeneration and wound healing effect.

2) DNA Synthesis Salvage Pathway

Cell growth was accompanied by internalization of PDRN-derived nucleotides to offer purine and pyrimidine rings for the “salvage pathway”. In fact, damaged of hypoxic tissue very often cannot undergo to the DNA “de novo” synthesis. Under these conditions, salvage pathways operate to recover bases and nucleosides generated from the breakdown of DNA and RNA. The salvaged bases can then be transformed into nucleotides and reincorporated into DNA. PDRN generates nucleotides and nucleosides that can contribute to DNA formation, offering a faster tissue regeneration and wound healing effect.

Efficacy Experiments
※ The mechanism of action of PN is almost the same as PDRN.

1) Anti-Aging

(1) Cell regeneration: PDRN promotes the growth of human dermal fibroblasts.

(2) Collagen regeneration: Proline is one of the most highly represented amino acids in collagens. The cells cultured in the presence of PDRN enhance collagen synthesis and secretion into the medium.

(3) Anti-oxidation: Anti-oxidative activity was assessed by DPPH assay. PDRN showed antioxidant activities in a dose-dependent manner, and those activities can suppress cellular oxidative stress in skin cells.

2) Repair

(1) Anti-inflammation: PDRN can effectively decrease the inflammatory cell counts, indicating its anti-inflammatory effect.

(2) Repair of ultraviolet B (UVB) damage: Cyclobutane pyrimidine dimers (CPD) is a typical indicator UVB damage. PDRN can effectively reduce CPD percentage, proving its repairing effect on UVB damage.

(3) Wound healing : In the in vitro scratch assay, a significant increase in percentage of wound closure was observed in the PDRN group compared with either the control group or the IL-1β group, suggesting PDRN repairing effect on wound healing
